Detection of DSB Repair Foci as a Potent Tool in Biodosimetry and Cancer Research
Radiobiology currently experiences its renaissance, especially in light of the globally increased threat of terrorist attacks, potential industrial disasters, planned cosmic journeys, and (cancer) radiotherapy development. DNA double strand breaks (DSBs) represent the most serious DNA lesions induced by ionizing radiation and radiomimetic chemicals. In this lecture, we will focus on current possibilities of DSB detection and quantification by the means of H2AX immunofluorescence microscopy and flow cytometry. Applications of these techniques in biodosimetry and cancer research/medicine will be discussed. H2AX foci appear at sites of DSB in minutes after irradiation (as the consequence of H2AX histones phosphorylation on serine 139) and represent a specific, very sensitive DSB marker. The number of H2AX foci corresponds to that of DSBs and H2AX nuclear distribution is characteristic for high-LET and low-LET radiations.
